My protein gets precipitate in absence of urea during dialysis which makes it difficult to use this protein in affinity purification. Several non-ionic detergents, Triton X-100, NP-40, etc. are available but I'm not sure if they are strong enough to solubilize the protein. Any ideas if any combinations of several detergents can do the work?
Hello! Do you need to eliminate the detergent during dialysis, and also keep your protein in native form or soluble form? Do you want to perform an affinity purification after dialysis?
My area of expertise is mainly industrial cleaning so forgive me if my thinking is too far out of the box in your area; however, perhaps you could use something of a similar nature, such as potassium cocoyl hydrolyzed rice protein which is used as a protein surfactant for personal care products, or another similar material is potassium undecylenoyl hydrolyzed collagen (CAS 68951-92-8) which is a detergent. Common use is in personal care products and it reportedly has antifungal properties. This would be an approach similar to "like dissolves like" in assisting the solubility of your protein.
