Traditionally, according to the literature alloxan were used to induce diabetes mellitus in lab animals and then checking for the effects of some medicine or developed composites. Currently, I am looking for a natural product to induce diabetes in lab animal especially rat model instead of using a chemical.
Previously, I used egg yolk to induce hyperlipidemia instead of pure cholesterol in the rat model and I was very successful.

Streptozotocin can induce Type- 1 diabetes in animal model and Type- 2 diabetes with multiple low doses according to (Surgical Clinics of North America 81(3):527-42 · July 2001) and is a naturally occurring chemical.

Streptozotocin is natural, although it has to be extracted from the fungus strain and I think you want a plant or herb in toto. In that case, I do not know.

I remember that citrate buffer and its pH were critical in the success of the induction of diabetes. Sometimes you need a second injection. I used to have close to 100% success. Intraperitoneal, in rats.
