I think my former colleagues in Lyon, France, Nathalie Strazielle and Jean-François Ghersi-Egea, run a commercial service for their various BBB models, see p. 7 of the attached pdf.

I appreciate this will probably only help if you can ship out your molecules, but do check with them whether some of their systems can be taken to someone else's lab, too.

At the very least, as they are very knowledgeable, this should get you started!

You have a local expert there at Monash in Joe Nicolazzo!  Many use the immortalized human hCMEC/D3 cell line avail. from Coraud's group in France.  There are no commercialized products.  It certainly depends upon what phenotype you'd like to mimic as no in vitro model has a perfect in vivo phenotype.  Good luck.

I find these kits a little expensive, but if you have the founds for it, probably worth trying. 

I've used the BBB kit from PharmaCo-Cell Company Ltd. that Csilla mentioned above (Cat # PCC-RBT-24H) and found it very straight forward and relatively easy to use.  If you decide to use this kit then make sure you have an instrument to measure Trans Endothelial Electrical Resistance (TEER) like the EVOM2 and EndOhm-6 (World Precision Instruments).

The in vitro BBB models from Cecchelli's lab are well known and represent the most well-characterized and very tight in vitro BBB systems available, much tighter in term of permeability than HCMEC/D3 cells for example... some commercially available "ready to use" kits also exist in some species...really feel free to get in touch and check their literature as well. If you decide for TEER measurement, be aware that TEER values are apparatus dependent. You can only compare different tightness of existing in vitro BBB models using the same TEER system and  it depends of the type of inserts you use as well...so be very careful. If you rather decide for permeability coefficient measurement using an integrity marker, this is independent of any apparatus or any in vitro systems and much more reliable. HBMEC (Human Brain Microvascular Endothelial Cells) are a well-known and developed in vitro model for the brain endothelium (=blood-brain barrier). 

You could also consider a co-cultivation of HBMEC and astrocytes. The BBB is not only brain endothelial cells but astrocytes represent the supporting component of the blood vasculature. In my opinion, HBMEC+astrocytes represent the most complete in vitro system. The issue is to be able to culture them together, since they are two different types of cells. It might be worth to give it a try...good luck!

Yes, the xCELLigence system uses cellular impedance of attached cells to measure barrier function.  Grow an epithelial monolayer to confluence for several days and add your treatment and measure the loss of adhesion of the monolayer dynamically.  Visit www.aceabio.com and go to the Applications tab. There is an App Note for it as well as numerous user publications using this method. 

I recommend looking over the following paper: https://www.researchgate.net/publication/294105976_In_vitro_models_of_the_blood-brain_barrier_An_overview_of_commonly_used_brain_endothelial_cell_culture_models_and_guidelines_for_their_use.  There are no "ideal" in vitro models of the BBB endothelium in that the in vitro phenotype must be carefully examined for validity to the in vivo setting and that's question dependent.
