Dear Dr. Shaheen, Yes the use of HPLC for large number of samples on daily basis is not economical for you, so you can use alternative method, that is titration method, however this method isn't so accurate as HPLC method, but at least that method can reduce economically. In general by using the titration method , the amount of acid produced by lactobacillus spp. can be measured. By assumption that Lactobacillus spp. produced dominantly lactic acid, so the acid ampunt produced by Lactobacillus spp is estimated to be equal to the lactic acid amount produced by the same bacteria. sorry, it may be this method is a little bit help as alternative method.

i am already trying titration its really just give the rough idea about the amount produced by bacillus spp. but not accurate enough to adopt it in standard research work. is spectroscopy helpful to estimate lactic acid by using standard solution?
Actually the accurate method for estimate lactic acid bacteria is HPLC method, However, Its better you try one sample to estimate lactic acid by using spectroscopy with standard solution and HPLC. If the result of lactic acid estimate by using spetroscopy will nearly close to that by HPLC method, you can use spectroscopy with standard solution for a lot of samples to be estimated its lactic acid. Hopefully, this matter is a little bit help to check the method as alternative method.
