A 16s amplicon sequencing from mixed bacterial-algal culture system showed moderate rhizobium reads ( abundant bacteria - 55%, other moderate bacterial reads ranged from 2% - 30%, Rhizobium species- 10%). When plated on agar using different media 2216E, RO, modified 2216E, Humic enriched media and rhizobium media from Himedia M408 couldn't grow rhizobium colonies. It might be due to competition from other bacteria.
Can anyone suggest a better media or technique/tips by which i might possibly isolate this species?
Note: i need to isolate it as i need to further study it's role in the system.

You can use YMA (Yeast Mannitol Agar) media with specific antibiotic treatment.

Well, did not find any luck with Graham's selective media. Though, YMA with congo red (without antibiotics) yielded few different species (not rhizobium) which i did not find with general media. The problem is rhizobium holds just a small part in my complex bacterial culture. I will try to modify antibiotics dosage and see if i can get it. Thanks
