Hi, Can anybody tell me is there any difference between these two proteins? I am doing western blotting for expression analysis of MDR1 protein. So I am using MDR1 monoclonal antibodies. Predicted molecular weight is 130-180 kDa, but i got two bands done higher molecular weight above 250 and one lower below 77kDa.
In the specification of Recombinant Anti-P Glycoprotein antibody i read that it detects a predominant protein band migrating in the region of 180-200 kDa and typically will demonstrate a smear on the membrane in the region of the 150 – 300 kDa due to the glycosylation profile of the protein.
Abcam recommends not boiling the sample after lysis. No such specification found incase of MDR1 antibody. So I am confused either i can use p-glycoprotein antibody instead of MDR1 and then follow their advise? or may i treat sample without heating with same MDR1 antibody and see result?
