I want to observe the dynamics of sperm count in human bodies. I need a method that allow multiple tests in a human being in one day. Non-invasive way better.The masturbation method may not be a good idea because everytime you get a data of a time point, you have to wait the sperms to be generated or released in to epididymides. Is there such a method?
This method, if there is, can be a very useful technique to investigate the male reproductive physiology and toxicology. It may also help to interpret the large intra-individual variation of male semen parameters.
Thank you for you answers, Dr. Al-Delemi and Dr. Kadhim! However, what I need is a technique by which I could investigate the semen quality of one man at different time points in the same day. I am not sure whether these papers supplied such a technique. It seems not feasible to collect semen from the same man several times a day, nor to get testis specimen from a man several times.

Sperm count. The sperm count is made when the specimen has become fluid. Mix the specimen thoroughly and dilute it 1:20 with a fixative solution containing 5 g sodium bicarbonate, 1 ml formalin, and 99.9 ml of dH2O. This dilution may be made by pipetting 0.1 ml of semen and 1.9 ml of the diluting fluid. Let the mixture stand until the mucus dissolves. Shake it thoroughly and load a hemacytometer. Count the spermatozoa in the same manner as you would count RBCs. The calculations are as follows:
Sperm count x 20 (dilution) x 5 (area) x 10 (depth factor) = sperm/rnm^3
Then convert your answer to sperm/ml by multiplyingthe above answer by 1,000. Example: 155 x 20 x 5 x 10 = 155,000 sperm/mm3
155,000 x 1,000 = 155,000,000 sperm/ml
Normally, a cubic millimeter of semen contains 60-150 million spermatozoa
