Hi, I was wondering how to determine the "sample height" as it is called at the Dionex or "injection height" at Agilent LC-instruments when you have very little sample volume and already use a vial with inlet. You could try it and lower the height until either the vial has a hole in the bottom or the injection needle is broken/bent, but I wanted to avoid that. Does anyone here have any experience with this?
I don't have an idea how to adjust the height of the needle; however, I suggest you use vials with a small sample capacity to raise the height of the solution indirectly.
In my humble opinion, it is suggested that it is prepared different vials with various volume of analyte and then considering that the type and dimentsions of septum and needle diameter of syringe different tests were done. Of course, doing such an experiments is usually expensive and and comes with high costs. In fact, the most of all is that the analyte should be entered to column inlet of apparatus even it is too little.

The option you propose is quite risky, as it does not take into account any possible fluctuations in the thickness of the bottom of the micro-inserts, or the possibility of using inserts from another manufacturer.
It seems to me that injection of a larger volume of a diluted sample would be a simpler and more reliable solution, if your samples can of course be diluted.
