I'm interested in fixing and imaging cells after staining their microtubules (live, of course), so I'm looking for a microtubule stain for which fluorescence signal is not lost after fixation.
For TEM, yes. Lead citrate and U.A. and Osmium staining should all indicate the microtubules. Lead and U.A. staining occurs post sectioning of resin-embedded material, while OsO4 staining occurs as part of the fixation process of the tissue.I would recommend the use of all three for a given tissue.
Hi Leelabati Biswas , You can use MitoView mitochondrial dyes from Biotium. Even though they said it’s for live cell. You can use the methanol based fixation with this dye.
