I am not sure if I do understand your question. However, the quality of the image by the camera depends of the quality of your objective, if you have an apochromatic obj. 1000x with a high resolution camera, your images will be better than a plan apo obj. Now some microscopes can multiply the magnification to 2.6x. but the resolution is not good. There are software that help cleaning and improving the contrast. if you want more resolution on an microscopic level, consider confocal microscopes or SEM's .

Dear Dr. Ronald Ochoa , my concern is to capture the image of the mite specimen at maximum magnification possible. Currently, with all sources available (built-in microscope + camera attached) I am only able to get the magnification of x200. I am curious if I can take images (avoiding to use SEMs) at a better/higher resolution of some characters like empodia or striae or setae etc. I highly appreciate your reply and thank you very much.

Dear Jawwad (Alsalamo Alikum) - The Olympus magnification changer you have (U-CA) provides magnification increments of 1.25x, 1.6x, and 2x. Therefore, your highest magnification power should be calculated as follows: 10x eyepiece + 100x oil obj. + 2x mag. changer = 2000x. Using 20x eyepiece will increase your mag. more, but keep in mind that the max. USEFUL magnification power of a light microscope is 1,500x. If you need more, as Ron suggested, try SEM which can magnify images up to 200,000x.

I share the opinion of Ronald Ochoa. To get better picture you need to have objectives of high quality. If you have N-plan (this is mentioned on the objective) you can do it with semi-apochromatic or apochromatic ones. For a given magnification these lenses have a better resolution, less chromatic aberration etc (apochromatic is better than semi-apo but more expensive), therefore they give you a more detailed picture.

As said by Mohamed Negm a magnification changer multiply magnification, it only increases the size of your picture without increasing its resolution (sometimes useful to draw).

To observe and draw empodia, lobes on dorsal striations etc. I use oil immersion objectives x100 semi-apochromatic (by Leica) coupled with x10 eyepieces (magnification x1000 up to x2000 with magnification changer).

To improve the quality of your observations do not forget to check the settings of your microscope like the basic centering of light, Köhler illumination and Bertrand lens adjustment.

Observing specimens through eyepiece surely gives me a higher magnification but I can not take images from the eyepiece.

I want to take an image of the mite specimen through a camera (attached to the photo port of the microscope). Here I need C-mount adaptors or T-mount adaptors. Can I increase the magnification of the final image captured? Please help me with this issue.

Dear Jawwad - In my opinion, conventional magnification most acarologists use is enough. Because, as you know, magnification power inversely proportional to image resolution. If you are able to add DIC, it will be a fantastic addition to your machine. All the best - Mohamed

I am not aware of any such device.

If you use a microscope camera use one that has a very high resolution. This will allow you to digitally enlarge (a little) without losing any detail of the final captured image.

As an example, in the laboratory, we use a camera attached to the photo port of the microscope (c-mount) with a 18 mega pixel sensor. This allows me to make drawings (empodium, knob of aedeagus etc.) directly from the screen using a pen tablet.

Dear Dr. Philippe Auger its a great suggestion by the way. We are currently in the process of attaching a DSLR camera with the microscope. I think as you mentioned, it is the best way around.

Dear Dr. Mohamed W. Negm thank you again for the suggestion about DIC. In our lab, as you know better than me, we have interference contrast accessories but they are neither strictly DIC nor we have been able to correctly use them. But from now, I will focus more on DIC acc. to your suggestion.
