is there a live cell stain that is NON-florescent?
I can do trypan blue stain for dead, and an MTT for live cell metabolism, but I'm ideally looking for a stain like trypan blue but only for LIVE cells. I don't have access to a florescence compatible microscope and the only thing I have to read absorbency in a plate reader (aka lysed cells). Any suggestions welcome. Thanks.
You do not need always to lyse cells to measure viabilty. You can measure protein content by SRB method. You can measure LDH after lysis - this will show live cell signal if you first change the old medium to a proper buffer. MTT is what you use, does it what you need (if you accept metabolic activity as live cell signal). There are soluble MTT analogs with less hand-work to detect metabolic activity. XTT (or Alamar Blue) is one of, i think you can detect both an absorbance or a fluo signal of it and you just read your plate after adding the compound and incubate it.
