I want to measure the bioactivity of epo with a bioassay. I am thinking of using TF1 (a human erythroleukemia cell line) cells, but do I use CCK8 (faster, non cytotoxic) or MTT (more literature)? Or something else?
What do you think?

Both CCK8 and MTT assays are commonly used to measure cell viability and proliferation, and either one can potentially be used to measure the bioactivity of EPO on TF1 cells.
CCK8 assay is based on the ability of viable cells to convert a water-soluble tetrazolium salt into a formazan product, which can be measured spectrophotometrically. It is a faster and more sensitive assay than MTT, as it requires a shorter incubation time and generates a stronger signal. CCK8 assay is also non-cytotoxic and can be used to measure cell viability in real-time.
MTT assay, on the other hand, is based on the reduction of MTT by mitochondrial dehydrogenases in viable cells to form insoluble formazan crystals, which can be dissolved and measured spectrophotometrically. This assay is commonly used in the literature and has been well validated.
Both assays have advantages and disadvantages, and the choice between them may depend on factors such as the specific research question, the experimental design, and the resources available. If you are interested in measuring the effects of EPO on cell proliferation or viability over a short period of time, CCK8 assay may be a good choice. If you want to evaluate the long-term effects of EPO treatment on TF1 cells, MTT assay may be more appropriate.
It is also worth noting that other assays, such as flow cytometry or ELISA, can be used to measure different aspects of cell biology and may be more appropriate depending on the specific research question.
