After preservation of the strains of pseudomonas aeroginosa for a period of 4 months at -20 ° C, I transferred the strains on a nutrient agar medium for the manipulators thereafter. After incubation for 2-3 days at 30 ° C, the strains do not show the pseudomonas-specific blue-green color. My question is, is the pigmentation of pseudomonas aeroginosa can be influenced by the temperature and / or duration of the conservation? Or are there other factors that can influence pigment production?
it is very much dependant upon temperature...the entire growth curve changes, also oxygen depletion can effect the pigment production.
Pyocynin is the blue color pigment produced by P. aeruginosa. It is extra cellular virulence factor produced by strain to cause cellular lysis in infected host host. The production of blue pigment by the strain is influenced by time, temperature, media composition,etc in vitro condition. I have extracted it in late log phage and estimated in various condition. Under the treatment of LPS exctrated from Marine bacterium the pyocynin production was reduced by 40% in my experiment.

The production of Pyocynin by the strain is influenced by in vitro conditions. However if you try to re-cultivate your strain again at earlier conditions of the isolation, the pigment will return back.

I think this depends on your storage conditions. Under stress conditions, it is possible that some virulence genes of this bacterium to disappear. This may happen if glycerol is not added enough. It is also best to test other virulence genes.
