Hello,
I am trying to measure the inflammation levels in HLA-B27 transgenic rats which get colitis. I measured plasma TNF-a levels and they are low. I have measured them in portal vein blood's plasma though. Does anyone know if what happens with cytokine levels of portal vein blood? Are they expected to be high in an inflamed rat?

We have never measured TNF in portal vein blood, it is expected that the concentration will be same. TNF is an early cytokine of the immune system and hence the plasma level tends to decrease in the aggressive phase of the disease. You can detect TNF in colon tissue using protocol provided in the following link.
