We are new to the estradiol field and trying to measure levels in female rat serum. The kit I currently have (Arbor Assays Serum EIA Estradiol kit ) only measures unbound estradiol.

Will the unbound portion of estradiol fluctuate/cycle along with the total amount in circulation?

Which would we report in a paper? The unbound fraction only?

It is unclear if the "normal" levels of estradiol reported in papers are bound/unbound or both.

We could also measure the levels in urine if this provides a more reliable measure.

There appears to be a lot of speculation about the reliability of estradiol EIA measurements, but we do not have easy access to LC-MS or the funds to support that. We are looking for some guidance about how to proceed.
