I need an Antibody for detecting HSP70 B2 (An isoform of HSP70) in Crassostrea gigas (Pacific oyster). I ordered MAB3846 (mouse Ab) from Millipore. Its datasheet says that ''the epitope of clone 3A3 is located between amino acids 504-617 of human Hsp 70, a region that has been shown to be involved in stress-induced nucleolar localization''. I tried to blast the HSP70 B2 from mouse and C. gigas and found that there was a stretch of 12 conserved amino acids and several other smaller stretches of 3-4 amino acids in the region between 504-617 (Please have a look on the attached file for the clear picture). Will it be a suitable Ab for this study?
Just give it a try, but do not get disappointed if you do net the required result. This is because HSP70 and HSP70 B2 are not totally different antigens.There may not be similarity but some homology has been reported. The question is whether these homologous sequences are in the concerned epitopes.

Yes, HSP 70 and HSP70 B2 are very similar and I understand that an Ab for HSP70 can possibly work for HSP 70B2 as well.But the Ab I have ordered is for mouse (as I cant find it for oysters or other bivalves or fishes). According to datasheet the epitope lies in the region 504-617. But the BLAST output shows a conserved region of only 12 amino acids. And I doubt if this long stretch (only 12 amino acids long) is enough to be identified by the Ab.
I agree, ultimate answer lies in trying it.....I'm quiet anxious now :-'(
