I want to observe the bioluminescence in cells which result from Luciferase, while microscope with CCD camera is unvalilable in my instisute. So Is it possible to view the bioluminescence via confocal microscopy (Leica TCS SP5)?
Only if you fit the CCD or preferably an EMCCD camera onto the confocal microscope. Then the camera needs to be run at -80c with an exposure time of 10 min (for EMCCD) or 30 min (for backthinned CCD). The confocal microscope also needs to be switched off completely and in a very dark room.
