Am working in the research topic of non enzymatic electrochemical sensor for Glucose detection. I have a doubt on it is possible to use two electrode system which means (working electrode and counter electrode i.e, pt) to take the reading? This thing is right or wrong? Please give me your suggestions... Thanks in advance...

Yes u can Use
We can use either two electrode or three electrode system
In cyclic voltammetry we vary the working electrode potential over a range with respect reference electrode whose potential is fixed
Function of each electrode as follows
WE (working electrode-Redox reaction of our interest occur and measured
CE-counter electrode-controlled by potentiostat to set WE potential and balance current
RE-Reference electrode -Electrode whose potential is fixed and provides WE potential feedback to the potentiostat.
When we use 2E system Working and counter (or reference electrode) i.e when counter and reference electrode is sorted with potentiostat.
In such case a single electrode performs both work.
When current is passed over the reference electrode (which act as both counter and reference) potential is not going to be constant and there by not going to act as reference electrode any more
Its preferable to use 3E SYTEM over 2E system

Thenmozhi it can be done with a two-electrode system; however, this type of electrode configuration can generally present some drawbacks because the "reference electrode" is also the counter electrode, there is not really a reference for the potential measurement due to the polarisation of the electrode. Therefore, the applied potential at the working electrode is not really controlled.
Some articles report on the use of two electrodes for biosensing with printed electrodes but I recommend that you use the three-electrode system if possible.
You can do this by short circuit connecting of reference and counter electrode cable terminal together to the electrode which is not involved in the catalytic reaction. And, the working electrode terminal from the instrument to an electrode where the interest of the catalytic reaction (sensing transducer).
