The tissue sections that I mostly use are from fresh frozen organs usually without any fixation. The thickness is around 10-12 microns. I routinely use H&E staining, but is now in need of a better contrast for the nuclei. It would be of great help if anyone has a protocol/suggestion for the same and is willing to share? Any help would be highly acknowledged.
It is not a good idea because of the eosin can give the strong signal too. It is better not to add other staining except of DAPI but you can use one specimen and make several slides for DAPI and for H&E separatedly and then compare.
Thanks for all your answers! @Morea: Yes I need an alternative for Hx for nuclei (one that gives a high contrast image suitable for detecting the nuclei by any optical analysis) along with a counterstain that helps me to visualize and interpret the morphology of the corresponding tissue. Let me know if you want to know anything else.
