It should still work if you've stored it in the fridge, it didn't dry out and you kept it clean. HRP is fairly robust: you've probably stored that secondary antibody in the fridge for years now (before adding it to the blot).

Assuming you left the membrane in the 4 degree celsius fridge and the membrane didn't dry out, then just adding fresh ECL and developing will still allow you to detect your protein. You do not need to strip and reprobe.

Thanks for all your answers. Unfortunately, I stored my membrane at room temperature, dry, between 2 whatman sheets. I can try to put freshh II Ab and then develop again, but not sure whether this will work.
