Perhaps by using a fluorescently tagged peptide/aptamer that specifically binds to the gene of interest (accompanied by noticeable change in emission spectrum / intensity / shift upon successful binding).
It'd be much appreciated if relevant reference is included.

Hi there,
PCR using gene specific primer set or restriction digest analysis will give you the information but you will need sequencing to check the construct anyway.
