I'm doing antibacterial activity assay on several compounds. The problem arose when my compounds only dissolved in 100 % DMSO. I worried if DMSO will affect the diameter of inhibition zone. Would you like to inform or suggest me, what should i do ? Thanks
As long as the DMSO is diluted to a level that does not significantly affect the bacterial growth, then it is OK to make a concentrated stock solution in DMSO. The final DMSO concentration to which the bacteria are exposed should be no more than a few percent. You could do a test to see how sensitive your bacteria are to DMSO itself in order to set the upper limit on DMSO concentration.
Usually, people take less than 10% DMSO concentration for the antimicrobial activity as it is found in some studies that conc. less than 10 does not have any effect but in your case, you can take 100% DMSO as a negative control so that you can correlate your result, whether it is your compound giving any activity or it is the DMSO.
Adam B Shapiro thank you for your answer, I already dissolved my compounds using 5% and 10% DMSO, but they didn't form homogenous solution.

I think you can try to use a negative control of 100% DMSO and DMSO at the concentration used in dissolving the test compound. In principle, the negative control here is used to dissolve the compound, not to inhibit the enzyme activity, so that the effect of DMSO on the test compound that will be used in the enzyme inhibition test can be estimated.

just Include control wells containing only the bacterial culture and the highest concentration of DMSO you plan to use in your assay. This will tell you if DMSO itself has any inhibitory effect on bacterial growth.
