In the seismic analysis of a water tank, the acceleration time history is applied in horizontal direction. Is it necessary to include acceleration due to gravity in vertical direction along with the time history in horizontal direction? is there anything wrong in considering so? If I do the analysis considering vertical acceleration along with horizontal component, shall I need to add it to "g"?
To my understanding the g effect is already considered if you are considering the self weight of the structure.
You have to perform gravity analysis either prior to dynamic analysis or both together to simulate initial state of stress. In case of water in a tank, its depends on how you include hydrodynamic pressure in your analysis.

I have given density (in kg/m3) as input. It will take into account in the formation of mass matrix.
But my question is, whether adding "g" into the inertial force (in Z direction) along with seismic acceleration ( X direction ) in will create any effect on the transient analysis of the structure

As you mentioned the modeling of the fluid to be done by Fluid 30 elements. you can consider the gravity loads from self-weight of the structure at the nodal points ( in Z direction) and can run both static and dynamic analysis under this load. However, If your fluid element is not considering the hydro-static pressure then you may need to consider the self weight of water as well. I am not sure about that.
