Recently I am reading literatures about Fast Green staining, and get confused: Fast Green is used in Trichrome staining to label collagen fiber, but sometimes it is also used together with Picrosirius Red to stain non-collagen part. May I ask whether it stains collagen or not, please? Or it binds collagen according to pH/Concentration?
Hello Guang Li, I have attached a page out of J.A. Kiernan's book "Histological & Histochmical Mathods: Theory&Practice, 2nd Edition, Paramount Press. I hope it will help you to understand how these stainings are working. My own expierence: The original van Gieson's stain with acid fuchsin and picric acid und the Masson-Goldner Trichrome stain don't use heteropolyacids like phosphormolybdic acid (PMA) or phosphortungstic acid (PTA). Without these acids you can stain collagene tissue very specific (van Gieson red; Masson-Goldner blue wirh aniline blue or green with fast green).
