Hello, anybody knows if drug delivery using nanoparticles affects kinetics? Needless to say that I have to perform the experiment, as different factors (type of drug, nanoparticle, cell line - PCa) will affect... But I am curious if anybody has experienced an early or delay response when using nanoparticles.

Could you please be more specific? What kind of nanoparticles and drug? What are we comparing the kinetics to?

Even without any experience, one could think that particles labelled with molecules and naked molecules would diffuse differently. Although the name is nano, they are not freely diffuse in the cell or ECM or tissues unlike naked molecules that may dissolve and diffuse easily in aqueous environments. Yes, it will be definitely delayed or efficacy will be quite low unless you increase nanoparticle concentration.

First of all, You have to take into account the size of the particles and its porouosity. If they are more, the kinetics of drug release will be slower. Other case, nanoparticles are the object of cell phagocytosys and could give some other interaction with cells than the drug has been accepted by diffusion. Returning to nanoparticles in solution, check their leve of agregation. It depends of their surface charge. The molecule of the drug could change its conformation in interaction with nanoparticles. In my opinion yes. All the factors you mentioned could effect the kinetics of nanoparticles. I suggest you to take care your carrier materials and drug physicochemical properties and most importantly the binding efficiency between these two.
