We are looking into fibrogenesis of primary hepatic stellate cells (human) in vitro. We used TGF-beta as fibrogenic stimulus and among other parameters we found increased PDGF-mRNA expression. Up to now I did not find any data on PDGF production or maybe even secretion by HSC, yet (while of course PDGF does stimulate fibroblast cell types). Could it be that HSC may secrete PDGF in an autocrine fashion?
Is it the alpha chain that you detect or the beta chain ? Because I found in an article that fibroblast do secrete the alpha chain of PDGF but not the beta one (Table 4 ; http://onlinelibrary.wiley.com/store/10.1046/j.1463-1326.1999.00032.x/asset/j.1463-1326.1999.00032.x.pdf?v=1&t=htbr19w0&s=c14e654f62dac956bcd7a8b2e3b979b0bedf0ccf)

PDGF A-chain mRNA has been detected in activated human stellate cells . During liver injury, stellate cells display increased PDGF production as well as the upregulation of PDGF receptors .
