I am crossing Camk2a-Cre (from Jax) and floxed mice with td-Tomato gene.
- I tried both female and male Cre drivers as I am concerned about germ line recombination.
Note: FF = homozygous, FW = heterozygous, WW = wildtype
- Genotyping results don't follow Mendelian genetics ( FF mouse x FW mouse generated some WW pups.)
- As you might have known, Camk2a-Cre is forebrain specific driver, but I even get "pink pups" (pink ear/limbs)- with non specific expression of reporter gene.
- The most weird thing I've encountered so far is red fluoresced pancreatic islets obtained from Cre NEGATIVE FF mice. According to qPCR, our gene of interest wasn't deleted in islets (again it was Cre NEGATIVE + Cre shouldn't be active in peripheral organ).
It'll be amazing if anyone could give me some ideas of what it is going on here. Also, any suggestions for articles would be helpful.
