After Skin Prick Test and Oral Food Challenge, What would be the best method or choice of a researcher among these:
1. Immunoblotting (Western blotting, dot blotting),
2. Immunoassay (e.g. ELISA, RIA, CAP)
3. The Cellular stimulation test (e.g. BAT)
4. Combination of these

the techniques you have mentioned are all usable, the only limitation concerns the ratio between IgG and IgE that there may be in the serum to be analyzed: IgG directed against the allergen are often more numerous than IgE. it may default to the technique used.
If the question is to study the connection between IgE and allergen, you must clone the B cell that produces the desired IgE.
You have different studies which show the performance of using the CD40 system (or HuBBB) in targets such as BetVI or DerPI; publications that combine Denepoux, Lebecque, Valenta, Visco ...
On our side we are currently working on peanuts being followed by ELISA after cloning B cells, it gives very good results
It depends on what your research objective is. In case you want to prove clinical relevance of SPT determined sensitization DBPCOFC is the golden standard and you do not need anything else. Otherwise you can use any of the mentioned for different investigative purposes.

One question for you. Food extract is used when perform SPT. According to my understanding, extract may contains multiple food allergens e.g. in hazelnut different allergen molecules have been identified (Cor a 1, 2, 6, 8, 9,10,11, 12,13, and 14). Then how we can diagnose that which allergen molecule is causing allergic reaction? Or SPT and food challenge only identify the problematic food instead of allergen molecule?

What an excellent point! By SPT and DBPCOFC you ove sensitization to food allergens and its clinical relevance. It is very important to know the molecular profile of a sensitization to food or inhalant allergens. In the former case it is important for dietary regimen choice and has a great prognostic and predictive value. In the latter you can discriminate between true, co- and cross sensitization and choose the right allergen for AIT. So the answer is - you have to use molecular allergy diagnostics. Thermo Fisher UniCAP method with molecular allergen components and ISAC micro chip technique are the golden standard. You have to bear in mind that many companies have developed immunoblot ELISA methods for detecting sIgE to allergen components. Check i.e. Euroimmun, Germany, Luebeck Euroline Single Purified Allergen Component IgE tests for pollen, insect venom and food allergens.
