Hello all, please provide some information if you can. I am trying to achieve a high throughput process of extracting BAC DNA. I need this for NGS so it cannot have genomic DNA or RNA present. Right? I currently have a system for extracting relatively quickly. However, this method does not isolate BAC DNA from RNA and genomic DNA. Is there a cleanup method that anyone could suggest that would allow me to achieve purified BACs in a high throughput process (not Qiagen Maxiprep)? Or is there another kit or automated system that could achieve this?
This may not be high-throughput enough, but it does result in a clean exctraction: You can try PCR amplication or RE digest of your BAC (including only the sequence of interest for extraction) then separation by gel electrophorsis.
