Recently, I did my HeLa cell transfection with a nucleus localization protein and then immunostained the cell with DAPI and specific antibodys, I found that the cell with relatively higher expression shows global chromatin compaction and the nucleus seems to be "broken", just like the photo showing. What's the cause of this phenomenon?Apoptosis or necrosis? Could anybody tell me, thank you

Dear Cai Liang,
The cells in which you are having more chromatin compaction are the cells who have just entered into mitosis (prophase). This kind of condensed chromatin you will see after Nuclear envelop breakdown at the onset of mitosis. You can use mitotic marker-pH3 (pS10) to confirm the same. Hope this may be helpful to you.
