We want to measure only the mature form of IL-1beta, not the pro-form in mouse lung, but all the ELISAs I've looked at measure both (or can't be sure whether or not they measure the pro-form). Can anyone suggest something that only measures the mature form?
This isn't an exact answer to your question, but you could consider supplementing your ELISA results with WB data. For example, measure the IL1b by ELISA, and then concentrate the sample and run it by WB to probe for IL1b. In a clean system, you should only (or vastly predominantly) detect mature form in culture supernatants or even in most animal serums. For example, Figure 1A in the linked paper. If you can show that you get elevated ELISA results, AND that your WBs show that in your system only mature form is detected in concentrated samples, then you can be fairly confident that your ELISA data is only for mature form.
