Hello everyone,

I am screening for Inhibitors. Km of my enzyme is 20uM.

but at 20uM substrate conc I am not getting a good signal.

The graph curves very quick and also the signal to noise ratio is around 1.5 only. Z´is very good around 0.8-0.85.

But if I increase the substrate conc. to 30 or 40uM. I get a good initial reaction (where I could stop the reaction and read the output). Also the signal to noise ratio increases to 2.1-2.6.

Z´is also good around 0.8.

But I am wondering will I miss a potential Inhibitor? if I screen at 30 or 40uM ATP (Km is 20uM).
