I have 25 mL of silver nanoparticles bought from sigma aldrich to check the invitro anticancer activity in A549 cell lines . specifications of the nanoparticle. are 20 nm particle size (TEM), 0.02 mg/mL in aqueous buffer, contains sodium citrate as stabilizer . I want to know in which solvent it can be dispersed so that i can treat the cell line or can i use the nanoparticle directly. someone please suggest me a solution for this.
Dilute (dispersion implies that they're not fully dispersed when you buy them and I doubt this) in the same continuous phase that they are supplied in. You can recover this by centrifugtion (reatining some original material for dilution) or the recipe may be stated on the MSDS.

If your confusion is how to dilute the AgNP you have for the cell treatment, the solution is going to be the medium you use to grow A549 lung cancer cells. if i am not wrong it is DMEM, so you have to dilute the AgNP with the FBS containing DMEM.

Almost certainly if you change the ionic make up of the nanoparticle from the medium that they were originally in, then irreversible aggregation and agglomeration will probably result.

You can prepare the suspension of silver nanoparticles by adding NPs to distilled water (pH 7.0) containing sodium dodecylbenzene sulfonate (SDBS, 0.1 mM) to increase the dispersity of the NPs followed by 1 hr of ultrasonification (40kHz, 250 W).

You'll also contaminate the sample with the probe material formed by erosion/cavitation.
