I am about to work with biotic stress on Mung Bean and French Bean seed varieties. There is no tissue culture room, so how can I maintain optimum in- vitro growth condition in our laboratory?
You should try with the sterile medium. I think its better to use vermiculites or sterile sands for germination, after all you dont need nutrients for seed germination. And the advantage of using sterile medium like vermiculites or sand instead of potting mixture is that you wont get any soil borne diseases........I guess this works
Soaking in water for 12-18 hrs is required for germination as the seed coat of beans seed is very hard, then seeds can be placed in dark for 2-3 days at 25-28C with adequate moisture (95-100% RH).
Soaking in water makes the seed coat soft and congenial for germination, hence no cutting of seed coat is needed. However, you may consult any book of seed technology, which has a chapter on germination.
