In my research, I developed a biphasic polymer scaffold for bone tissue engineering. Given that the scaffold consists of two functional layers, I would like to directly place the bone marrow aspirate on the scaffold to test different cells' migration or function. Would anyone provide some suggestion on the in vitro cell (cells in bone marrow aspirate) tracking? Thanks in advance.

How long would you like to track the cells for? Do you have access to a confocal or fluorescent microscope?
I have used CellTracker dyes to track osteoblast-like cells in 3D constructs. These dyes are available in multiple colours, and they are stable for up to 72 hours, non-toxic, and quite bright. I hope this helps!

In vitro studies, Isolation and culture of cells and preparation of implants, cell attachment and viability assessment are most important part of your study.
First you to find out whether there are dead cells or not? To characterize it, it is advice to use SEM to know swelling ratio, significant difference in cell proliferation among cells cultured with control medium and different concentrations of scaffold extracts at the same culture time, fluorescent labeling etc.. Most importantly, whether residual cells remaining in the scaffold or not? Live cells distributed in the scaffold with dead cells or without dead cells etc.? SEM can confirm that seeded cells could anchor onto the scaffold, live cells were distributed in the scaffold, with no dead cells (if it is red fluorescence) being found.
