No ,it is important to have knowledge of their biological variability. I have different coefciient of variation for reticulocytes a)Normal CV:5-8% b9 Low count 2-4 % and c) High count CV 10-14% .I ask What was the coefficient of variation according to the counts that were conducted in this stuidio?

Howell-Jolly bodies, basophilic stippling as well as other intracellular inclusions can interfer in automated reticulocytes count.  The magnitude of overestimation depend on staining method (methylene blue, fluorescence [oxazyne, acridine orange, other]) and on methodology/technology systems.In the cited Etienne Mahe anc Coll. article none of the 100 samples has morphologic evidences (microscopic or CellaVision) of intracellular inclusions.

The imprecision of all the methods decreases as the reticulocyte concentration increased  .All methods at all concentrations shows an imprecision less than that of the microscopic  .The CV is a máximum  of 15 % for  all concentrations . It has an imprecision less than that of the microscopic method.  The need for low imprecision in tests used for reticulocytopenic patients can be  in monitoring the early erythropoietic response after bone marrow transplantation that presents a advantage:  the possibility of knowing the imprecision at every concentration   

As well described by Juan Carlos, the automatic and microscopic reticulocytes count is affected by an high inaccuracy and imprecision. These analytical problems increase at lower concentrations. For these reasons the reticulocytes enumeration cannot well describe the productive response after bone marrow transplantation. The immature reticulocyte's fraction (IRF) or their hemoglobin content and concentration or MCVr are better descriptors of effective and early erythropoiesis
