When different solvents are used for extracting phenolic contents from different sources, should gallic acid be dissolved in corresponding solvents for standard curve. Else could gallic acid solution be prepared using methanol as solvent? When gallic acid is dissolved in methanol or water due to difference in solubility the slope value of standard curve are too different. So , this leads to tremendous difference in mg GAE/g dry sample.
first, dry your extracts and then dissolve them in methanol. use gallic acid standard dissolved in methanol.

I recommend you use methanol to make your standard curve with gallic acid (GA), I wouldn’t use water because it makes much difficult to dissolve the GA and can affect your curve. Regarding the solvents used for the extractions, I suggest making a blank for each solvent when you are performing your TPC assay.
Do it with water. It will give you accurate std graph. Methanol will not give such a graph. When you go for methanol, it will have a lesser R2 value compared to water. Also, you might face the problem of precipitate when you add Na2CO3 if you use Methanol instead of water, which would interfere with your absorbance values.
