I'm doing some cFos immunohistochemistry, and wanted to check the general consensus on reporting the results.

My predecessor worked out the count/area (microns), and scaled this up to count/mm2. Howver, looking at the literature, many groups looking at similar IHC only report the total count.

What do people think about this? Personally, I don't see how count alone is robust enough, without some compensation for variation in nuclei area - I know myself that the exact size and position of the amygdala varies from animal to animal, and can be different from that shown in Watson & Paxinos due to the angle of the section cut.
