The AM ester form is essentially non-fluorescent and does not have an appreciable affinity for calcium. The main problem that you would encounter is that the cell will continue to accumulate Fura-2. However, I do not anticipate that this would be a serious problem for a ratiometric dye, but you should take the precaution of recording the Ca2+ insensitive fluorescence more frequently than would otherwise be necessary.
