We are trying to do an immunofluorescence stain of cells grown on coverslips using MYH11 antibody (Santa Cruz). Usually we have no problems with this technique, but this antibody is giving us a hard time. Did anyone ever work with it? Any insights will be appreciated
We never used MYH Abs from Santa Cruz. Good Abs to myosin II isoforms were from Covance. Fixation by 1%PFA in serum-free medium 15 min, followed by cold Methanol.
Vera, if this "specific" antibody causes headaches, I would go for another company. Since I personally did not work with this particular antibody and other researchers need to know what host you would prefer, I would recommend you this website for now. http://www.biocompare.com/Antibodies/ You can browse for reactivity, host, application, conjugates, you name it. I have a great protocol if you intend to amplify your signal by biotinylation, using streptavidin-conjugates for visualization.
