If the samples are fresh pollen (i.e. not sediment samples or something like that), they can often be identified satisfactorily without any special treatment. This depends on having a good reference collection, some experience with pollen microscopy, and tolerance for uncertainty with some taxa (e.g. species within Rosaceae). The slides can be made more or less permanent by the addition of phenol (very toxic!) to the mounting medium, but I have made slides with simple glycerin jelly (no phenol) and sealed with clear nail polish that showed no signs of degradation over the two-year period during which I was using them.

You could use alcohol (methanol or ethanol) if you do not mind the bleaching effect it has on the pollen.

I have not tried this method out myself, but Dr. Jen O'Keefe did some work using an enzyme-based method.
