I think (and hope) that you made calibration lines with known concentrations of MB. If you did, there is no problem; just read the concentrations. Calculate degradation (CMB0 -CMBt)/CMB0 * 100, and you have the % degradated.

Why is 2 different WLs used? The peak for MB is around 660 mn, far away from the UV region.

A UV lamp has very low, if any, energy at 660 nm, where MB has the highest absorption. But for sure, there must be Abs even in the UV range.
