if you wanted to add to PLA, you could try immuno-TEM (we did in recent circ res paper). label two proteins with different size gold beads, view by EM then use a programme (e.g. MetaMorph) to measure distance between the two beads - in theory you can measure the distance in the nm scale a little closer than 40nm, but it may be subjective as compared to PLA, but could allow you some more insight. For my studies it showed very tight associations with the proteins and anything over 20nm was not considered interaction. (we also backed up with PLA). Also ours was looking at effects of PTM (phosphorylation) also
