I want to analyse a particular protein cd24's role in chemo resistance, and I have found out with MTT assay that shCD24 cells shows more death than wild type cancer cells for 2 of the chemotherapeutic drugs. so If I wanna analysis the combinatorial effect of both the drugs, can I just mix the IC50 concentrations of both the drugs and will do the 24,48,72 hours time points ? is that fine?

I recommend you to do the following steps:
1. Determine the IC50 for each drug individually. I advise you to use graph Pad Prism software to analyze the results and finding the IC50.
2. Prepare mixtures from both of drugs with different ratios. You can do it by using one of the drugs with a constant consentration.
3. Select the best ratio of drugs with the maximum cytotoxicity on cancer cells.
4. Prepare mixtures of drugs with different consentrations and with the best and unique ratio of them determined in step3.
5. Determine the IC50 for combination of drugs according to the results obtained from step4.
6. Use the formula developed by Dr. Ting Chao Chou to evaluate the synergy between the drugs by obtaining the combination index from three IC50 you measured.
