If I am working with plant extracts and their photoprotective properties.... It is just enough to evaluate the citotoxicity at a specific (high) concentration of my extracts or it is necessary to determine the IC50 of each extract?...In order to determine the security of the use of the extracts... I am going to use human dermal fibroblasts.
The use of dermal cell models is good for phototoxicity experiment but the whole point of cytoprotective assay is to find a drug that protect cells from external damage without itself causing any toxicity. Thus, you do need to determine the toxicity profile of your drugs ideally through determination of IC50 value. Your test should be based on the concentration of your compounds that do not at all cause any sign of toxicity to the cells. This concentrations would need to be determined from the IC50 value study instead of try and error with unknown safe concentration.
