I am working on the valorization of cocoa pod husks powder into bioproducts. I would like to estimate lignin degradation products after alkaline delignification. I think by determining the total phenolic components I could get an idea of the extent of delignification. I would also like to have the calculation formula and the procedure. Thank you so much.
Procedure for estimation of Phenols and polyphenols by Amino-antipyrine method. Ettinger, M., Ruchhoft, C., Lishka, R. Sensitive 4-Aminoantipyrine Method for Phenolic Compounds. Anal. Chem.1951, 23, 12, 1783-1788.
Reagents:
1. Phenol standard: (0.1mg/ml): dissolve 10mg of crystalline phenol in 0.1NHCl. To dissolve completely heat it to 60oC on water bath.
2. 4-amino-antipyrine: 1mg/ml solution
3. 0.5N sodium hydroxide
4. 0.5M sodium bicarbonate
8. Potassium ferricyanide: 5mg/ml in distilled water
Procedure:
1. Add varying volumes of phenol standard (0.2, 0.4, 0.6, 0.8 and 1 ml), take distilled water as blank or 0.1 ml of sample-nanoparticle (Do not forget to equalize the volume for all tubes in standard)
2. Add 0.8ml of 0.5N sodium hydroxide and 1.2 ml of 0.5M sodium bicarbonate. (Shake the tubes)
3. Add 1 ml of 4-amino-antipyrine reagent (Shake the tubes)
4. Add 1 ml of potassium ferricyanide (Shake the tubes)
Take the reading at 520nm in a colorimeter or spectrophotometer.
