okay, so I decided to add PEG 2000 to my liposome formulation. should I add PEG 2000 by itself or should it conjugated with ones of the lipids I have used. the only issue is it will be way more expensive.
What is the need to add PEG 2000? The addition of extra surfactants will make the shell of the micelle more stable and concrete. This thereby will be difficult to release the contents entrapped inside. If PEG itself is the cargo, then you need not to conjugate with the lipid. Just dissolve with the organic solvent along with lipids.
It would be better to use modified lipid monomers such as DSPE-PEG-2000 and/or covalently conjugate PEG to liposomal monomers in vitro rather than simply physical absorption of PEG on lipid monomers.
