Dear sir, in our lab we generally practiced extracting DNA from Vibrio bacteria is the "boiling method" the simplest, cheapest & effective method for extraction. Especially when you have lots of Vibrio isolates need to screen them in that case if you use a kit for extraction that will be quite expensive.
For specific molecular work, the CITAB DNA extraction method will be cheap & effective.
"boiling followed by immediate cooling is the easier method for initial screening, After that CTAB method is the one of the effective method for extraction.
2) Inoculate in any enrich broth (I will use BHI broth) and incubate for overnight.
3) Go for Phenol Chloroform Isoamyl alcohol procedure for DNA extraction. This will give maximum yield but unpurified DNA. Machery-Nagel commercial kit is available for this procedure. You can purchase and go with the kit also or conventional method is also ok.
