I want to take extract of Estrogen from leafy vegetables. What you think about methodology? If I use ultrasonic solvent extraction then which solvent is good for recovery? Some scientists use the acetone, potassium phosphate etc. But I don’t know which solvent provide the maximum recovery. If you have any experience about this please share with me. If you can send me all steps of methodology then it will be very helpful for me. Please keep in mind after this extract I want to run my sample on GC-MS.

First of all you extract of your sample using solvent of different polarity. then isolate the compound in pure form using TLC/Column chromatography /HPLC to check the purity of your compound. Then you can run GC-MS, IR, NMR of your compound.
Estrogens in plants are usually classified as phytosterols and are freely soluble in non polar solvents like hexane or chloroform you can prepare one of these extracts and then separate estrogens using column chromatography controlled by TLC analysis using one of standard phytoestrogens as reference
Prepare plants containing Estrogens extract in EtOH.
Dry it to remove EtOH traces and then dissolve in chloroform, filter and dry it again.
Now use this extract for GC-MS analysis.
