I think, you may use the method of Frankenberger and Tabatabai (1980a) using formamide (CH3NO) as substrate for determination of amidase activity. The final detection of ammonia nitrogen produced by the enzymes activity was performed by distillation with 25% NaOH and titration with H2SO4 (0.0005M). The attached files may also help you for other methods of amidase estimation.

yes of course, but I can't to get this chemical reagent so I thought others posibilities to use diferent chemical reagent...and overcome that problem..
